Un equipo de investigadores dise\u00f1\u00f3 mediante herramientas computacionales un <strong data-start=\"513\" data-end=\"535\">candidato a vacuna<\/strong> contra <em data-start=\"543\" data-end=\"571\">Renibacterium salmoninarum<\/em>, bacteria responsable de esta patolog\u00eda, dando un primer paso hacia el desarrollo de una vacuna basada en m\u00faltiples prote\u00ednas y regiones antig\u00e9nicas del pat\u00f3geno.<\/p>\n<p data-start=\"774\" data-end=\"1309\">El trabajo, publicado en la revista <em data-start=\"810\" data-end=\"860\">Frontiers in Cellular and Infection Microbiology<\/em>, emple\u00f3 un enfoque completamente <strong data-start=\"894\" data-end=\"907\">in silico<\/strong>, que combin\u00f3 gen\u00f3mica comparativa, vacunolog\u00eda reversa, inmunoinform\u00e1tica, modelamiento estructural, acoplamiento molecular y simulaciones de din\u00e1mica molecular para identificar los componentes bacterianos con mayor potencial inmunol\u00f3gico. Los resultados corresponden a <strong>predicciones computacionales<\/strong> y constituyen una base para futuras validaciones experimentales.<\/p>\n<p data-start=\"1311\" data-end=\"1806\">La investigaci\u00f3n surge en un contexto en que la BKD contin\u00faa representando un importante desaf\u00edo sanitario para la industria. De acuerdo con datos oficiales citados por los autores, durante 2024 la enfermedad explic\u00f3 el 9,2% de las mortalidades por pat\u00f3genos en salm\u00f3n Atl\u00e1ntico, el 12,1% en salm\u00f3n coho y el 8,4% en trucha arco\u00edris. Adem\u00e1s, concentr\u00f3 el 51,5% del uso de antimicrobianos asociado a enfermedades durante la etapa de producci\u00f3n en agua dulce.<\/p>\n<p data-start=\"1311\" data-end=\"1806\"><strong data-start=\"112\" data-end=\"163\">El estudio fue desarrollado por Jeffrey Araneda<\/strong>, del <strong data-start=\"169\" data-end=\"220\">Laboratorio de Gen\u00e9tica e Inmunolog\u00eda Molecular<\/strong> del <strong data-start=\"225\" data-end=\"336\" data-is-only-node=\"\">Instituto de Biolog\u00eda de la Facultad de Ciencias de la Pontificia Universidad Cat\u00f3lica de Valpara\u00edso (PUCV)<\/strong>; <strong data-start=\"338\" data-end=\"368\">Patricio A. Flores-Herrera<\/strong>, del mismo laboratorio y del <strong data-start=\"398\" data-end=\"475\">Centro GEMA \u2013 Gen\u00f3mica, Ecolog\u00eda y Medio Ambiente de la Universidad Mayor<\/strong>; <strong data-start=\"477\" data-end=\"494\">Waldo Acevedo<\/strong>, del <strong data-start=\"500\" data-end=\"599\">Laboratorio de Qu\u00edmica Biol\u00f3gica del Instituto de Qu\u00edmica de la Facultad de Ciencias de la PUCV<\/strong> y del <strong data-start=\"606\" data-end=\"700\">Center for Interdisciplinary Research in Biomedicine, Biotechnology and Well-Being (CID3B)<\/strong> de la misma universidad; <strong data-start=\"726\" data-end=\"748\">Sergio H. Marshall<\/strong>, del Laboratorio de Gen\u00e9tica e Inmunolog\u00eda Molecular de la PUCV; y <strong data-start=\"816\" data-end=\"837\">Fernando A. G\u00f3mez<\/strong>, del mismo laboratorio.<\/p>\n<h3 data-section-id=\"kcsjf9\" data-start=\"1808\" data-end=\"1826\">Dise\u00f1o racional<\/h3>\n<p data-start=\"1828\" data-end=\"2389\">Como punto de partida, los investigadores compararon tres genomas completos de <em data-start=\"1907\" data-end=\"1924\">R. salmoninarum<\/em>, constatando un alto nivel de conservaci\u00f3n gen\u00e9tica entre las cepas disponibles. Sobre esa base analizaron aproximadamente 3.700 prote\u00ednas codificadas por el microorganismo e identificaron inicialmente 27 prote\u00ednas con potencial antig\u00e9nico. Tras una serie de filtros relacionados con su localizaci\u00f3n, antigenicidad y funci\u00f3n biol\u00f3gica, la selecci\u00f3n se redujo a 17 prote\u00ednas para continuar el desarrollo del candidato a vacuna.<\/p>\n<p data-start=\"2391\" data-end=\"2846\">Posteriormente, mediante herramientas de inmunoinform\u00e1tica, identificaron 154 ep\u00edtopos potenciales \u2014las regiones de las prote\u00ednas capaces de ser reconocidas por el sistema inmune\u2014, los que fueron depurados seg\u00fan criterios de antigenicidad, toxicidad y similitud con prote\u00ednas del salm\u00f3n. El proceso concluy\u00f3 con la selecci\u00f3n de 60 ep\u00edtopos agrupados en 26 regiones antig\u00e9nicas provenientes de 15 prote\u00ednas diferentes.<\/p>\n<p data-start=\"2848\" data-end=\"3201\">Con esos resultados, el equipo construy\u00f3 una <strong data-start=\"2893\" data-end=\"2946\">prote\u00edna quim\u00e9rica multiantig\u00e9nica y multiep\u00edtopo<\/strong> de 439 amino\u00e1cidos, que integra doce regiones antig\u00e9nicas derivadas de prote\u00ednas relacionadas con mecanismos de virulencia, sistemas de secreci\u00f3n, captaci\u00f3n de hierro, transporte de az\u00facares y chaperonas bacterianas.<\/p>\n<h3 data-section-id=\"5arc2g\" data-start=\"3203\" data-end=\"3236\">Resultados de las simulaciones<\/h3>\n<p data-start=\"3238\" data-end=\"3821\">Las simulaciones computacionales indicaron que la prote\u00edna dise\u00f1ada presenta estabilidad estructural, ausencia de toxicidad predicha y regiones antig\u00e9nicas expuestas que podr\u00edan facilitar su reconocimiento por el sistema inmune del salm\u00f3n Atl\u00e1ntico. Asimismo, los an\u00e1lisis mostraron una interacci\u00f3n favorable con mol\u00e9culas del complejo mayor de histocompatibilidad (MHC) de clase I y II, responsables de presentar ant\u00edgenos al sistema inmunol\u00f3gico, adem\u00e1s de mantener estabilidad durante una simulaci\u00f3n de din\u00e1mica molecular de 100 nanosegundos.<\/p>\n<p data-start=\"3823\" data-end=\"4106\"><strong data-start=\"3823\" data-end=\"4068\">Los autores se\u00f1alan que estos resultados proporcionan una base para futuras validaciones experimentales del candidato y respaldan el uso de metodolog\u00edas computacionales para apoyar el dise\u00f1o de nuevas estrategias de vacunaci\u00f3n contra la BKD.<\/strong><\/p>\n<h3 data-section-id=\"18z4hoq\" data-start=\"4108\" data-end=\"4123\">Pr\u00f3ximo paso<\/h3>\n<p data-start=\"4125\" data-end=\"4509\">Pese a los resultados obtenidos, los investigadores enfatizan que el trabajo constituye \u00fanicamente una etapa inicial del desarrollo. El estudio no evalu\u00f3 la capacidad del candidato para inducir protecci\u00f3n en peces ni reducir la mortalidad o el uso de antibi\u00f3ticos, ya que todas las evaluaciones fueron realizadas mediante modelos computacionales.<\/p>\n<p data-start=\"4511\" data-end=\"4924\">En ese contexto, se\u00f1alan que el siguiente paso ser\u00e1 validar experimentalmente el candidato mediante ensayos <strong data-start=\"4619\" data-end=\"4631\">in vitro<\/strong> e <strong data-start=\"4634\" data-end=\"4645\">in vivo<\/strong>, adem\u00e1s de evaluar su inmunogenicidad y capacidad protectora.
