Cada vez que el salm\u00f3n se infecta con <\/span>Piscirickettsia salmonis<\/span><\/i> -causante de la septicemia rickettsial salmon\u00eddea (SRS)-, dentro de sus c\u00e9lulas se desata una batalla silenciosa. En primera l\u00ednea est\u00e1n los macr\u00f3fagos, las c\u00e9lulas inmunes encargadas de atrapar y eliminar al invasor. Pero no siempre ganan: en ocasiones se da\u00f1an, se inflaman en exceso o incluso terminan sirviendo de refugio para la bacteria que deber\u00edan combatir.<\/span><\/p>\n

Esa contradicci\u00f3n es el punto de partida de la investigaci\u00f3n que desarrolla Juan Pablo Pontigo, acad\u00e9mico e investigador de la Universidad San Sebasti\u00e1n sede De la Patagonia, especialista en inmunolog\u00eda de peces. Su trabajo se centra en dilucidar los mecanismos intracelulares que se activan cuando los peces enfrentan pat\u00f3genos bacterianos y virales, una l\u00ednea que ha complementado con estudios en especies ant\u00e1rticas y su respuesta al cambio clim\u00e1tico, adem\u00e1s de la b\u00fasqueda de nuevas herramientas biotecnol\u00f3gicas aplicadas a la acuicultura.<\/span><\/p>\n

Un interruptor escondido en las mitocondrias<\/b><\/p>\n

Su estudio plantea que la clave para entender si un macr\u00f3fago de salm\u00f3n se repara, se autodestruye o permite que la bacteria sobreviva en su interior podr\u00eda estar en las mitocondrias. Aunque tradicionalmente se las conoce como las \u00abcentrales de energ\u00eda\u00bb de la c\u00e9lula \u201cestas estructuras tambi\u00e9n act\u00faan como sensores de peligro: cuando detectan estr\u00e9s o da\u00f1o, emiten se\u00f1ales de alarma que pueden orientar a la c\u00e9lula hacia la recuperaci\u00f3n o hacia una inflamaci\u00f3n destructiva\u201d, afirma.\u00a0<\/span><\/p>\n

El equipo de investigaci\u00f3n compara este mecanismo con un tablero el\u00e9ctrico dom\u00e9stico. Ante una falla menor, el sistema activa una se\u00f1al, corrige el problema y la casa sigue funcionando con normalidad. Pero si el cortocircuito se prolonga, el sistema se sobrecarga y el riesgo de incendio aumenta. Algo similar ocurrir\u00eda en los macr\u00f3fagos del salm\u00f3n: ese \u00abcortocircuito\u00bb mitocondrial podr\u00eda inclinar la balanza entre el control del da\u00f1o y una respuesta inflamatoria da\u00f1ina para el propio pez.<\/span><\/p>\n

Tres destinos posibles para una misma c\u00e9lula<\/b><\/p>\n

A partir de esta hip\u00f3tesis, la investigaci\u00f3n explora tres escenarios posibles para los macr\u00f3fagos infectados. El primero, es un camino protector en el que la c\u00e9lula detecta el da\u00f1o, se reorganiza internamente, controla la inflamaci\u00f3n y logra sobrevivir. El segundo, es un camino destructivo donde la se\u00f1al mitocondrial se intensifica, se activa el inflamasoma y la c\u00e9lula muere de forma inflamatoria, liberando nuevas se\u00f1ales de alarma. Y, el tercero, es quiz\u00e1s, el m\u00e1s preocupante desde el punto de vista sanitario: la c\u00e9lula no logra eliminar completamente a la bacteria y termina transform\u00e1ndose en un escondite que le permite persistir en el organismo.<\/span><\/p>\n

Para poner a prueba estos escenarios, el equipo trabajar\u00e1 con macr\u00f3fagos de salm\u00f3n en condiciones controladas de laboratorio, empleando modelos de da\u00f1o celular, infecci\u00f3n con <\/span>P. salmonis<\/span><\/i> y herramientas para medir el estado mitocondrial. La investigaci\u00f3n combinar\u00e1 microscop\u00eda, citometr\u00eda de flujo, separaci\u00f3n celular, an\u00e1lisis molecular y transcript\u00f3mica, junto con etapas de validaci\u00f3n directamente en salm\u00f3n Atl\u00e1ntico, lo que permitir\u00e1 conectar lo observado a nivel celular con lo que ocurre durante una infecci\u00f3n real.<\/span><\/p>\n

Cambiar el foco: de la bacteria a la respuesta celular<\/b><\/p>\n

La novedad de esta l\u00ednea de trabajo radica en desplazar la mirada desde la sola detecci\u00f3n de la bacteria hacia la comprensi\u00f3n de c\u00f3mo responde la c\u00e9lula que la enfrenta. Si el equipo logra identificar una firma mitocondrial capaz de indicar cu\u00e1ndo un macr\u00f3fago est\u00e1 repar\u00e1ndose, cu\u00e1ndo est\u00e1 entrando en una inflamaci\u00f3n destructiva y cu\u00e1ndo est\u00e1 permitiendo que la bacteria persista, se abrir\u00eda una nueva forma de anticipar el curso de la enfermedad, antes de que sus efectos sean visibles a nivel productivo.<\/span><\/p>\n

Los resultados esperados apuntan al desarrollo de biomarcadores tempranos de la enfermedad, plataformas para evaluar nuevos inmunomoduladores y estrategias orientadas a fortalecer la respuesta defensiva del pez sin favorecer la inflamaci\u00f3n ni la persistencia bacteriana.<\/span><\/p>\n

Un aporte a la salud de los peces y a la sostenibilidad de la acuicultura<\/b><\/p>\n

Comprender c\u00f3mo las se\u00f1ales asociadas al estado de las mitocondrias influyen en la capacidad del salm\u00f3n para controlar la infecci\u00f3n, limitar el da\u00f1o inflamatorio o favorecer inadvertidamente la persistencia de\u00a0<\/span>P. salmonis<\/span><\/i>\u00a0podr\u00eda aportar nuevas herramientas para abordar la salud de los peces en la industria.
